视黄酸对BALB/C小鼠骨髓瘤细胞增殖及6sDNA聚合酶的抑制作用  被引量:1

摘  要:本研究以终浓度33.3μmol/L的视黄酸和6.7μmol/L的阿糖胞苷与BALB/C小鼠骨髓瘤细胞株培养72小时,结果细胞增殖抑制率分别为45.3%和61.4%。以[3H]一dTTP参入法法测定BALB/C小鼠骨髓瘤细胞核纯化的6SDNA聚合酶活性,发现体外反应系统中加入终浓度为2μmol/L,4μmol/L,6μmol/L,8μmol/L,10μmol/L的视黄酸,6sDNA聚合酶活性的抑制率分别为12.4%,18%,25*%,30,7%和31.6%(除第1个剂量外,其余P<0.05~0.001),阳性对照终浓度为6μmol/L的阿糖胞苷的抑制率为49%(P<0.001)。并对视黄酸抑癌作用的机理进行了讨论。Mydoma cells from BALB/C mice were cultured with retinoic acid at.an end-concentration of 33.3μmol/L and with cytarabine at an end-concentration of 6.7μmol/L for 72 hours,the inhibitory rates of cell proliferation were 45.3%and 61.4%,respectively. The activity of the pure 6s DNA polymerase from myeloma cells nuclear of BALB/C mice was determined by the method of(3H)dTTP incorporation,retinoic acid at an end-concentration of 2,4,6,8 and 10μ mol/L inhibited the activity of 6s DNA polymerase in vitro with the inhibitory rates of 12.4%。18%,15.6%,30.7% nd 31.6%,respectively(P<0.05~0.001,except the dose uf 2μ mol/L),the inhibitory rate of Positive control cytarabine at an end-concentration of 6μ mol/L was 49%(P<0.001)The mechan hanism of antitumor action of retinoic acid was discussed.
