摘 要:我国的甲型肝炎病毒龙甲株,经免疫捕获病毒颗粒,再进行反转录和聚合酶链反应(PCR),获得结构基因区cDNA,用双脱氧法直接测定序列。结果表明,甲肝病毒在结构蛋白基因区有高度的保守性。将我国龙甲株病毒的结构区基因与甲肝病毒HM175野毒株和MBB作比较,发现其核苷酸变异率分别为4.5%和3.1%,氨基酸变异均为0.13%。从结构区基因各区段来看,核苷酸变异主要发生在VP1和PV3区域。he uncleotide sequence of the structural genes of LJ strain which was isolated from a chinese patient's stool specimen and had 6 passages in 2BS cell, was determined by the RT-PCR and direct dideoxynucleotide-base sequencing technique. Comparison of itsnucleotide sequence to those of HM175 and MBB strains shows highly conserved geneticrelatedness.The nucleotide divergence of Chinese LJ strain from HM175 and MBB isolates inthe structural region(including part of 5'noncoding region) was 4.5% and 3.1% respectively.The amino acid variation of LJ strain from HM175 and MBB was 0.13%,and 0.13% respectively.
