双氧水诱导淋巴母细胞株产生EB病毒相关抗原的研究

摘  要:研究了双氧水(H2O2)诱导淋巴母细胞株产生EB病毒相关抗原的作用条件,并观察了H2O2与丁酸钠和巴豆油联合作用于上述细胞后对EB病毒相关抗原的协同激活作用。结果表明,0.2mM的H2O2室温下处理B95─8细胞10分钟,培养5天后,其EB病毒壳抗原(EBV─VCA)阳性细胞率分别由4%和1%增至36%,结果与丁酸钠和巴豆油单独激活时无显著差异。证明H2O2可以代替具有致突或促癌的丁酸钠和巴豆油用于激活B95─8和P3HR─1细胞,但对P3HR─1EB─VCA和Raji细胞EBV早期抗原(EBV─EA)的激活不够理想。H2O2、丁酸钠和巴豆油联合作用于上述三种细胞可以获得最好的激活效果,B95─8和P3HR─1的阳性细胞率分别可达80%和64%,Raji细胞的EBV─EA阳性率可达14%。The optimal conditions for the EBV─associated antigen production of Iymphoblast cell lines induced by H2O2 and the synergetic effect of the concomitant use of H2O2,TPA and corton oil on expression of EBV─associated antigen gene of the Iymphoblasts were studied.The results showed that treatment of B95─8cells with H2O2 of0.2mmol/L at room temperature for 10 minutes and culture for 5 days could increase theEBV─VCA positivity rate from 4%to 36%.There was no significant difference betweenactivating B95─8 by H2O2 and activating it by TPA or by corton oil alone,and theferoreH2O2 can replace carcinogenic activation reagents,TPA and corton oil,for the acticationof B95─8 cells;but its effect on P3HR─1and Raji cells are not as good as that on B95─8cells.At the same conditions,however,the highest activation results can be achieved ifthe above three activation reagents are used simultaneously for activating each of thethree lymphoblast cell lines,with a EBV─VCA positivity rate of 80%for B95─8and 64%for P3HR─1,and with a EBV─EA positivity rate of 14%for Raji cells.
