摘 要:本实验通过在低压舱模拟高原的缺氧环境,研究了急性缺氧条件下,大鼠红细胞膜生物物理特性的变化。实验选择1,6-Diphenyl-1,3,5-hexatriene(DPH)荧光探剂标记膜脂质,反映膜脂微粘滞性,另外还观察了膜蛋白内源性荧光光谱的变化,结果显示:急性缺氧条件下,红细胞膜脂质有序性增加,流动性降低,在模拟4000m、6000m、8000m高度维持8h,膜脂流动性比平原对照显著降低;动物在6000m分别减压8、12和24h,发现随缺氧时间延长,流动性进一步降低。另外本研究显示,在急性缺氧条件下,红细胞膜蛋白内源性荧光光谱峰型及最大发射波长无明显改变,但荧光强度有所降低,碘化钾(KI)荧光淬灭效应也有一定差异。The changes of the biophysical properties of the cell membrane of erythrocytes during acute hypoxia were observed in rats.The fluorescence probe,DPH,was used to label the lipids of the cell membrane to determin its microviscosity and fluorescence spectra of the menbrane protein.It was found that under the conditions of acute hypoxia at the simulated altitude of 4000 m for 8 hours,there was an increase of the regularity of the membrane lipids of eryhtrocytes and the microviscosity was also increased significantly,When the rats were kept at a simulated altitude of 6000 m for 8 to 24 hours,the membrane fluidity was further decreased along with the prolongation of time.The map of interior fluorescece spectra of membrane proteins and the quenching effect of the fluorescence of potassium iodide suggested that the structure or the distribution of membrane proteins of erythrocytes could be changed under acute hypoxia.
