摘 要:本实验从佛山和成都两地的致倦库蚊群体中筛选出3个酯酶类型不同的有机磷(OP)抗性品系,FS-1、FS-2和CD-1品系。各品系的酯酶基因B扩增水平、酯酶活性和OP抗性水平三者一致。各品系酯酶B基因限制性酶切片段比较分析表明,我国的OP抗性库蚊群体中不仅有世界性分布的酯酶B1和B2,而且在佛山和成都地区各有一个新的独立扩增的酯酶B,分别命名为B6和B7。Three organophosphate resistant strains(FS-1, FS-2 and CD-1) were selected from collections of Culex quinquefasciatus from Foshan and Chengdu, China. Different esterase patterns were observed in the three strains. The numbers of esterase B gene copies were approximately 32-fold in FS-1, 8~16-fold in FS-2 and 2~4-fold in CD-1. The amplification levels of esterase B genes were correlated with esterase activities and OP resistances among the three strains.The restriction pattern comparison of the amplified region containing the esterase B gene revealed large differences among the three strains. The esterase B1 was found in FS-1 and two esterases, B2 and a new B haplotype (named B6), were detected in FS-2. Another new haplotype(named B7)was observed in CD-1. Esterase B6 and B7 had identical electrophoretic mobility, but had different restriction patterns within the regions flanking the esterase B gene, indicating that two distinct haplotypes were amplified independently in Culex populations in Foshan and Chengdu,China.
