摘 要:用葡萄球菌肠毒素A刺激体外培养的大鼠脾细胞所获得的γ-干扰素(IFN-γ),于接种约氏疟原虫子孢子前5h静注于Wistar大鼠体内,子孢子接种42h后肝内红外期(EEF)数量显著减少,其子孢子发育率仅为对照组的7%~20%,但EEF的大小及成熟度均未受影响。大鼠接种子孢子后2h给IFN-γ,则红外期的密度、子孢子发育率及红外期大小,成熟度均与对照组无明显差异。将子孢子与IFN-γ共孵40min,然后超速离心洗脱IFN-γ后再接种于大鼠,则红外期的密度、大小、成熟度也均类似于对照组。结果提示:IFN-γ的抗疟作用并非是针对子孢子或红外期本身,而是通过修饰枯否氏细胞活性,减少入侵肝细胞的子孢子数来表达的。Gamma-interferon(IFN-γ)was obtained through the culture of the splenocytes of Wistar rats and the culture was stimulated with the enterotoxin A of staphylococcus. Rats were injected with INF-γ 5 hours before sporozoite inoculation. It was found that 42 hours after inoculation,the density of exoerythrocytic forms(EEF)was markedly decreased and the viability of the sporozoites was noly 7%~20% of that of the control.However,the size and maturation rate of EEF were not affected.When IFN-γ was given 2 hours after sporozoite inoculation, the sporozoite viability,the size and maturation of EEF were all normal as compared with those of the control. When sporozoites were preincubated with IFN-γ for 40 minutes and then injected into rats,the density,size and maturation rate of EEF were not affected. The findings suggest that the anti-malarial effects of INF-γ are not directly act on the sporozoits or EEF but prevent the invasion of sporozoites through the modification of the activities of Kuppffer cells.
