摘  要:本文选择已生育子女的25~35岁正常男子的精液标本,分成正常对照及分别经洗必泰(2mg/ml)、NP-9(0.25mg/ml)、MN-863(0.2mg/ml)、Ⅲ号原药(0.05mg/ml)和复方Ⅰ号(0.1mg/ml)处理的六组。用组织化学、免疫组织化学、荧光染色等方法分别显示精子DNA、RNA、磷脂、糖类,SDH、LDH、AP、ATPase、hCG、IgM、Y小体等成分,比较用药前后的变化,从而评价上述几种外用杀精剂的作用与机制。实验结果表明此组外用杀精剂不损害人的精子细胞核及DNA、RNA、磷脂以及Y小体的形态。洗必泰可破坏精子质膜的脂糖蛋白,明显干扰精子的多种酶活性;MN-863、Ⅲ号原药、复方Ⅰ号可损害精子质膜功能,干扰用能酶的活性;NP-9可损伤精子质膜的脂糖蛋白,干扰其用能与供能酶的活性。这组人精子细胞化学定位的方法,可有效地检测人精子的缺陷与损伤。Sperm samples were collected from 3 normal fathers, aged 25~35 years old. Each sample was divided into six groups, ie the untreated and the 5 spermicides treated (chlorheridine 2mg/ml, MN-863 0.2mg/ml, Drug Ⅲ 0.05mg/ml, Ico 0.1mg/ml, NP-9 0.25 mg/ml). The composition variation of DNA, RNA, phosphatides, carbohydrates, SDH, LDH, AP, ATPase, hCG, IgM and Y-body in sperms were assessed by histochemical and cytochemical methods. Consequently the mode of action of those spermicides has been elucidated. They did not affect the morphology of the nuclei, plasmalemma and Y-boby. But chlorheridine would impair the glycolipoproteins of the sperm plasmalemma and inhibit several enzyme activities. MN-863, Drug Ⅲ, Ico and NP-9 would interfere the function of sperm plasmalemma and the activity of energy-dependent enzymes. Besides, NP-9 would further affect the energy-supplying enzymes. Evidently the cytochemial localization of fine structure of human sperm will help determining the defect and impairment of human sperm.
