摘 要:目的:探讨绝经后骨质疏松的预防及中西药物的治疗作用。方法:作者选用6月龄雌性Wister大鼠34只,随机分为:卵巢假切除组(n=9),卵巢切除十生理盐水组(n=9),卵巢切除十已烯雌酚组(n=7)和卵巢切除十甲蓉片组(n=9).并将以同种动物股骨、胫骨按Urist方法制备的脱钙骨基质在卵巢手术的同时,植入受试大鼠体内,术后第3日分组给药,用药6周。在用药的第23日和30日进行四环素标记。取右侧股骨及植入的脱钙骨,制备成不脱钙骨切片进行骨计量学测定。结果:卵巢切除组骨皮质厚度、骨小梁宽度、骨细胞陷窝长度和宽度及血碱性磷酸酶(ALP)等的改变与绝经后妇女骨质疏松的改变相似。同时脱钙骨的平均矿化沉积率、矿化延迟时间、诱导成骨量较卵巢假切除组脱钙骨各指标明显降低(P<0.01).雌激素和甲蓉片治疗组上述各指标明显增加(P<0.01).结论:甲蓉片和雌激素可促进骨矿化恢复骨再建过程的平衡,具有预防绝经后骨质疏松的作用。Objective : This study was undertaken for the prevention of postmenopausal osteoporosis and itstreatment with combined Chinese and Western medicine。 Methods : Thirty-four 6 month old female Wisterrats were divided randomly into four groups : 9 in the sham-OVX; 9 in the OVX + NS;7 in the OVX + DES(0. 0375 g/L)and 9 in the OVX + JRT( 1 tablet/ml). Demineralized bonematrices made, by using Uristmethod, from the femora and the tibiae of the same species of animal, were planted into the rats during theovariectomy. The medication to the four groups, as indicated above, began on the 3rd day after OVX andlasted for 6 weeks。 The labels of tetracycline were made on the 23rd and 30th day。 All rats were killed 6weeks after the operation, The right femora and the planted decalcified bones were processed undercalcifiedfor quantitative bone histomorphometry。 Results : In the OVX + NS group the mean wall thicknesses, themean trabecular widths, the bone cell lacunals lengths and widths all significantly reduced and the serum al-kaline phosphatose rose。 These were similar to the changes of osteoporosis in postmenopausal women。 More-over, in decalcified bone the mean mineralization deposition rats, the mineralization lag time and amount ofinduced bone were significantly lower than those in sham-OVX group. After treatment with DEX or JRT,the above indexed were increased remarkably。 Conclusions : It is suggested that JRT or DES can prevent thebone loss and increase the mineralization of bone in OVX rats by recovering the balance of bone remodeling。They can be used to prevent postmenopausal osteoporosis。
