摘 要:本文从精子超微结构和供能酶的角度,探讨Cu^(2+)影响精子运动的机制。结果表明1)大鼠附睾精子在Bakers′-Cu^(2+)液中置体外培育5′后,精子活动力即明显下降。2)Cu^(2+)抑制乳酸脱氢酶(LDH)、琥珀酸脱氢酶(SDH)的组织化学反应,其中以LDH活性的降低显著,并与精子活动力的降低呈平行关系。3)LDH同功酶PAGE示LDH-C_4相对活性降低。4)精子线粒体内呈电子致密的LDH酶反应颗粒明显减少且电子密度降低,而精子超微结构无明显改变。本实验提示: Cu^(2+)对精子LDH-C_4活性的抑制可能是影响精子运动的主要环节之一。30 samples of semens were collected from caudal epididymides of rats. 1ml of sperm suspension (sperm concentration: 1.0×10~7) was added to each experimental tube which contained 5ml of Baker's-Cu^(2+) solution (with Cu^(2+)1.38μg/ml) and to each control tube with 5ml Baker's solution only. All samples were incubated at 37℃. The sperm motility, lactate dehydrogenase (LDH) and succinic dehydrogenase (SDH) activities were observed by cytochemistry, electronmicroscopic cytochemistry methods st 5', 30' and 2h. Results indicated that, 1) LDH, SDH activities and sperm motility were all inhibited by Cu^(2+). But LDH activity decreased more rapidly than SDH. 2) The percentage of relative activity of LDH-C_4 tested by PAGE reduced and the LDH-C_4 activity in the inner mitochondril matrix decreased but no remarkable alterations in the ultrastructures of mitochondria and axoneme. The results made it possible to conclude that LDH-C_4 activity is inhibited by Cu^(2+) which may cause the loss of sperm motility.
