摘  要:从热带假丝酵母(Candida tropicalis)野生株1045通过HNO_2诱变,得到几株能利用C_(10)~C_(17)单~正烷烃及混合正烷烃(C_(10~14))产生与基质链长相应的α,ω—二羰酸的突变株SD_1、GU_(11)、LD_(12)、KT_(13)、C_1及YU_(?)这些突变株能生长在以正烷烃为唯一碳源的简单培养基中,无需添加非烃碳源。通过基质的流加.pH 值的调节,氧气的供给及种子浓度的选择等发酵条件的控制,在28℃下发酵5~6天,得到诸二羧酸的产量大多在80g/L 以上,最高达到131.9g/L,这个产量相当于野生株1045产酸量的41.22倍。产物经气相色谱分析,确认为与基质链长相应的二羧酸,纯度分别为96~98%。长期传代实验表明,这些突变株没有菌种退化现象,无需复壮便能得到产酸重复性很佳的结果,因而可以认为这些突变抹是产酸性能稳定的优良菌种。By treatment of the wild type,Candida tropicalis 1045,with nitrous acid,several mutants such as SD_1,GU_(11),LD_(12),KT_(13),C_1 and YU_m were obtainedwhich could produced a series of α,ω-dioic acids from alkanes of correspondingchain length (C_(10)~C_(17)).In the course of fermentation,by supplementation of substrates,regula-tion of pH value,control of oxygen and seeds concentration,all of thesemutants could produce 80 g/L dioic acids with one as high as 131.9 g/L whengrowing on a simple medium with alkanes as sole carbon sources at 28℃ after5~6 days fermentation.The highest yield was 41 fold of the original strain1045.The products were identified by gas-chromatograph.The range of puritywas 96~98%.The influence of n-alkanes chain length,starting concentrationof n-alkanes,substrate supplementation,pH value,etc.on the yield of dioicacids were investigated.These mutants were still strong and healthy without recovery after subcul-ture for years on malt extract-agar slant,the production of dioic acids werealso as high as usual.This study indicates that mutants SD_1,GU_(11),LD_(12),KT_(13),C_1 and YU_m described in this paper have the potential use as stable andhigh yield producers for commercial production of dioic acids
