胸腺五肽(TP─5)的固相合成及其对人血液若干生化指标的影响

摘  要:采用多肽固相合成法合成了胸腺生成素Ⅱ(Arg─lys─Asp─Val─Tyr─OH(TP─5)),经HPLC检查其纯度达98%以上,氨基酸组成分析与预期结果相一致,质谱法测得分子量与理论值相符合。自愿受试者肌肉注射TP─5(每次1mg/支,间日一次,共十次)后,其血液中超氧化物歧化酶(SOD)活性提高了175%;对血液中超氧阴离子自由基(O_ ̄(-)_2)的清除率达20%,并使红细胞免疫复合物花环率和红细胞C_3b受体花环率分别提高了26.3%和24.3%。Thymopoietin Ⅱ(32─36)OH,Arg─Lys─Asp─Val─Tyr─ON(TP─5)was synthesizedhy the solid phase peptide synthesis and BOC strategy.The purified peptide was judged to behigher than 98%in purity by using HPLC. The data obtained from amino acid analysis and fastatom bombing mass spectrum were in good agreement with theoretical values. Bioassays of blood samples which were obtained from volunteers after i.m. of TP─5(1mg/d. tertian injection,10 times),show the activity of SOD rose up to 175%,the rate of eliminationof superoxide radical(O ̄(-)_2)was around 20%,the rosette rate of RBC─C_36 and that of RBC─ICincreased 24.3/and 26.3% in blood samples separately.
