血清多巴胺的溶剂萃取与荧光分光光度法测定

摘　　要：用溶剂萃取法提取、荧光分光光度法测定血清多巴胺含量。激发波长和发射波长分别为λｅｘ＝３２０ｎｍ和λｅｍ＝３７５ｎｍ，线性范围０～３．２６×１Ｏ－６ｍｏｌ／Ｌ，回归方程Ｙ＝１．３０３６×１０－８Ｘ＋２．８２２４×１０－８，ｒ＝０．９９９８，最低检出限为１．１６×１０－８ｍｏｌ／Ｌ回收率为９７．７５％，批内和批间ＣＶ分别为４．２６％和６．５２％。本法简便、快速、稳定、经济，易于批量测定，适合临床应用。Dopamine in serum was isolated with solvent extraction system and determined with spectrophotofluorometry.The maximum excitation wavelength and maximum emission wavelength were at 320 nm and 375 nm,respectively.The following regression equation is found:Y=1.3036×10-8X+2.8224×10-8,r=0.9998.The relative fluorescence intensity is proportional to the dopamine concentration in the range of 0￣3.26×10-6mol/L.The minimum detection limit was 1.16×10-8mol/L.The average recovery rate was 97.75%.Within-run CV and between-run CV were 4.26% and 6.52%,respectively.This method is found to be simpler,quicker,steadier and more economical than others.It is suggested that the spectrophotofluorometric method is applicable to the clinical determination of dopamine in serum.
