国产依丽黄酮抗去势大鼠骨质疏松的实验研究

摘　　要：切除３月龄雌性大鼠双侧卵巢后，口服国产依丽黄酮１２周，与单纯卵巢切除组对照，用骨计量学等方法对国产依丽黄酮药效及剂量反应关系进行综合评价。结果显示：切除卵巢后骨量显著减少，骨无机质含量降低，破骨及成骨活性均增加，但破骨大于成骨；口服依丽黄酮后，各剂量组恢复至接近正常水平。由此表明。In order to evaluate the effect of Ipri- flavone( BO,synthesized for the first time by theSchool of Pharmaceutical Sciences of WCUMS)against osteoporosis in ovariectomized(OVX)rats,98female Wistar rats were divided into 7 groups: bilat-eral OVX,sham operated(S),and 5 treatmentgroups after OVX, of which 3 groups were given do-mestic BO-25,100,and 400 mg/kg BW(L,M,H),respectively, and the remaining two groups, BO(made in Japan)-100 mg/kg BW(J) and 17β-estradi-ol-4 mg/kg BW (E ).One day after operation,BOand E were given daily and twice a week. respectively,by gavage to the animals,while S and OVX groupswere treated with vehicles alone,On the 14th and 6thday prior to the sacrifice 0.25%tetracycline hy-drochloride was administered i.p. to 6 rats of eachgroup at a dosage of 25 mg/kg BW. The rats weresacrificed after 12 wks and blood samples were collect-ed for serum Ca,P,and AkP activity. The uterineswere removed and weighed. Undecalcified sections ofthe left proximal tibia were processed for bone histo-morphometry,and HE sections of the right tibia wereprocessed for bone pathology.The weights per unitbone length, the calcium and phosphorus contents ofthe left femur were meaxured. The results showed that in the OVX animals tra-becular bone volume,trabecular thickness,and theash content of bone were all significantly reduced.This bone loss was associated with increase in osteo-clast index ,osteoblast index ,osteoid surface and serum AkP activity. The distances between tetracy-cline double labelings were also markedly elevated. Allof the above indices exhibited increase in boneturnover, and that bone resorption was much more inexcess than bone formation.In contrast , administra-tion of the BO to OVX rats resulted in the nor-maliza-tion of trabecular bone mass.The bone mass of Mgroup recovered nearly to the condition of the S groupand E group.A marked inhibition of bone resorptionwas observed, and uterine weight increased only in theH gronp.
