摘 要:本文检测糖尿病大鼠肾小球产生的转化生长因子β(TGFβ)的水平,并探讨体外高糖环境对系膜细胞产生TGFβ的影响。建立STZ诱发的糖尿病大鼠模型,4周后肾小球分泌的总量及活性TCFβ均比对照组明显增加,其中活性TGFβ增加更为明显,体外高糖培养后的系膜细胞产生的总TGFβ明显下降,而活性TGFβ却有上升。结果表明高糖环境下肾小球和系膜细胞确实存在着TGFβ分泌和激活的异常,TGFβ可能参与了糖尿病肾病系膜区扩张的形成。lterations in the synthesis and activities of polypeptide growth factors havebeen hypothesized to be involved in the mesangial expansion during diabetic neph-ropathy. This study was designed to examine the secretion of TGFβby both glo-meruli from diabetic rats and cultured mesangial cells under high glucose condi-tions. STZ-induced diabetic rats were made.TGFβlevels in the supernatants obtained from isolated glomeruli 4 wks after injection were determined by using a standard bioassy system. The results showed that both total and active TGFpβsecreted byglomeruli from diabetic rats were shgnificantly increased as compared with control .The increment of active TGFβ is more significant. Mesangial cells pre-cultured un-der high glucose conditions exhibited decreased secretion of total TGFβ, while in-creased production of active TGFβ,These results suggest that abnormal production and activation of TGFβexist in the glomerulus and mesangial cells under highglucose conditions. TGFβmay be involved in the development of mesangial expan -sion during diabetic nephropathy。
