摘 要:用嗜肝病毒动物模型对磷甲酸三钠的抗乙肝病毒作用机理和效果作了初步的研究。18只持续感染鸭乙肝病毒的麻鸭腹腔内隔日注射200~400mg/kg磷甲酸三钠共6周,给药期间分别取血清和肝组织以斑点杂交和Southernblot试验检测乙肝病毒DNA、结果显示:尽管用药过程中血清病毒DNA滴度有一过性下降,但肝内仍显示出活跃的病毒复制。表明磷甲酸三钠的抗乙肝病毒作用机理主要不是在于抑制病毒DNA或RNA聚合酶,而且它的抗乙肝病毒效果也并不理想。The antiviral effect and mechanism of Foscarnet have been studied with a hepadna viridae animalmodel. 18 Chinese ducks persistently infected with duck hepatitis B virus were given Foscarnet intraperitoneally,200mg to 400mg/kg every two days for 6 weeks. Serum of duck hepatitis B virus DNA and intrahepatic replicative forms of the virus were measured by dot and Southern blot test.We still found active viral replication in the liver,although there was a transient decrease of serum level ofduck hepatitis B virus DNA during the treatment.So, we conclude that the antiviral mechanism of Foscarnet is not mainly on the inhibition of viral DNA or RNApolymerases and the anti-duck hepatitis B virus effect of the drug is an unsatisfactory way of treatment.
