我国肠道传播戊型肝炎的病原形态学研究

摘　　要：采用简易免疫电镜方法，从我国新疆非甲非乙型（戊型，ＨＥ）肝炎病人粪悬液中检出含有小圆形病毒颗粒的阳性标本５例，经细胞培养，从接种含病毒的２ＢＳ细胞中分离到一株能引起细胞病变、具有血凝活性的病毒（８７Ａ株）。电镜下可见感染细胞胞浆中呈现局灶性空泡、液泡病变区，并含晶格样排列的圆形病毒颗粒，核变形。该株病毒能被新疆疫区以及来自缅甸、印度、前苏联ＨＥ病人血清及美国接种ＨＥ病毒的黑猩猩急性期血浆所聚集，显示其间有密切血清学关系，提示该株病毒为我国新疆ＨＥ的致病病原。该病毒大小约３０ｎｍ，圆形，表面形态结构特殊；装配成熟过程与粗面内质网、包涵体、微丝等密切相关；病毒因细胞破碎而释放。这些形态及形态发生特征与嵌杯病毒科病毒相近。我们认为应归嵌杯病毒科新属成员。For detection of the etiological agent of non-A, non-B hepatitis (HE), we have collectedstool specimens from 48 patients with acute hepatitis in XinjiaW, China. Virus-like particles were foundin the specimens of five patients by immune electron microscopy. Specimens with virus-like particles wereinoculated into human embryo lung diploid cell culture. A virus called strain 87A was isolated. Infectedcells showed obvious cytopathic effect. A small area of proliferation of vesicles could be found in the cyto-plasm of infected cells at 24 hours afrer inoculation , and later smooth membrane bound vesicles frequentlyassociated with paracrystalline arrays of virions could be observed. The virions were roughly spherical,and were 27nm to 34nm in diameter. The viral particles in the infected culture suspensions and purifiedvirions were specifically aggregated with sera of patients with HE from our and three other countries , andwith plasma of chimpanzee acutely infected by HEV, but not with those of patients infected with HAV orHBV and normal human serum. The results of IEM suggested that the strain 87A virus was the etiologi-cal agent of non-A, non-B hepatitis in Xinjiang, China. The morphology and morphogenesis of the strain87A virus reveal that it may be a new member of Caliciviridae.
