摘  要:采田间患病小麦叶片,经PEG沉淀、20%蔗糖垫(含0.3%TritonX-100)离心和氯化铯平衡密度梯度离心获得提纯病毒。紫外扫描呈典型核蛋白吸收曲线,A260:A280为1.15。病毒粒子呈线条状,长度为200~2000nm。病毒核酸为两个组分。免疫电镜观察,小麦梭条斑花叶病毒(WSSMV)抗血清和小麦黄花叶病毒(WYMV)抗血清均能强烈的“修饰”该病毒。提纯病毒免疫家兔,所得病毒抗血清效价可达1/1024,ELISA检测病毒灵敏度达0.128μg·mL-1。The virus was purified from infected wheat leaves by precipitating with PEG followed by differential contrifugation with 20%sucrose cushion comtaining 0.3%Triton X-100 and centrifuged in CsClisopycnic gradient.The uItraviolet absorption spectra of the purified virus preparation was typical of a filamen-tous virus and the A260:A280 ratio was 1.15.The particle length distribution of purfied virus was from 200 to2000 nm. It has two species of single-stranded RNAs and single coat protein with MWs of 28 kD. In ISEMtests,the virus particals were not only trapped effectively from saps but also decorated strongly by both WSS-MV-antisera and WYMV-antisera. Antiserum having a titer 1/1024 in micropreceptin test was obtainedfrom rabbits which had been immunized with the purified virus.The detection limition of the virus bv ELISAwas 0.128 μg/mL.
