摘 要:探讨了四种不同配比(6:3:1;4.5:4.5:1;3:6:1;5:5:0)的乙醇:丙酮:水混合液提取叶绿素 Chlorophyll(Chl)的方法(混合液提取法),并与 Arnon 法进行了比较研究。结果表明:四种不同配比的混合液在室温(13-18℃)下暗处直接浸提的玉米和小麦叶片的 Chl 提取液与同等材料的 Arnon 法 Chl 提取液有一致的吸收高峰,Chl 含量可用Arnon 公式计算;不同配比的混合液提取的 Chl 含量基本相等,且均高于 Arnon 提取法;提取液在暗处室温下存放5—7d 后,Chl 含量下降不明显,18d 后有明显的下降。混合液提取的Chl 在弱光(玉米1214Lx,小麦1450Lx)下4.5—5.5h,其含量下降极显著,但幅度均小于Arnon 提取法。由此证明混合液提取法具有 Arnon 法的主要特点,且在某些方面优于后者。The methods with four mixtures of alcohol,acentone and distillwater in- different proportions of 6:3:1,4.5:4.5:1,3:6:1 and 5:5:0 for extraction of chlorophyll(mixture methods)were conducted and subsequently compared with Arnon' s method.The results showed that the chlorophyll suspension extracted from Zea mays Land Triticum aestivum leaves with the mixtures had the same absorption peak as that with Arnon's method at the room temperature(13-18℃)and the chlorophyll contents could be calculated by Arnon's Formula;the chlorophyll contents obtained by the mix- ture methods were basically equal and higher than that Arnon's method,and the con- tents did not decrease significantly after 5-7 days but decreased significantly after 18 days in dark.The chlorophyll contents extracted with the mixtures decreased significant- ly at 1% level after 4.5-5.5 hours in weak light(maize 1214 Lx,wheat1450Lx),but the decreasing amount were less than that with Arnon,s method,We concluded that the mixture method had the main benefits of Arnon's method and was superior to Arnon's method some aspects.
