摘  要:用52种RAPD引物对橡胶树11个抗白粉病品系和11个感病品系基因组DNA进行RAPD分析,找到了一个和橡胶树抗白粉病表型密切相关的DNA片段,此片段长390bp,命名为opv—390。opv—390为11个抗性品系所共有和11个感病品系所没有。用ECL(enhanced cheminesence)酶标法标记抗性品系RRIC52(CK)的OPV390为探针,经Southern blot表明,该序列在6个抗性品系中均为单一拷贝或低拷贝,而感病品系皆不含此序列,由此看来,OPV—390可作为橡胶树抗白粉病基因紧密连锁的RAPD标记。这为今后钓取橡胶树抗白粉病基因的工作打下了基础。The genomic DNA isolated from 11 mildew ( O,idium hevea )-resistant and 11 susceptible clones of Hevea brasilliensis were amplified with 52 random primers by RAPD technique, A DNA fragment named OPV-300, which is closely related to mildew resistance phenotype, was detected in 11 resistant clone, Hybridization of restriction fragments of DNA from all resistant and susceptible clones with the probe prepared with OPV-300 amplified from RRTC 52 ( CK ) genomic DNA indicates that OPV-300 was a single copy or low copy sequence in resistant clones. By interence, OPV-300 may be a part or flank sequence of mildew-resistance gene. This may result in a possibility for seperation of mildew-resistance gene, from Hevea tree.
