香石竹脉斑驳病毒（CarVMV）鉴定和抗血清制备

摘　　要：将纯化鉴定后的香石竹脉斑驳病毒繁殖在美国石竹上，样品匀浆，澄清，两轮聚乙二醇沉淀后，硼酸缓冲液重新悬浮．即为粗提纯的病毒制剂。紫外光谱吸收测定，Ａ２６０～２９０／２８０为１．１２，制剂纯度７１％，病毒含量２．７５ｍｇ／ｍｌ。肌肉注射家兔得到微量沉淀效价１：１０２４的抗血清。这是国内首次制备出香石竹脉斑驳病毒抗血清。Diseased leaves of Dianthus barbatus inoculated with carnation vein mottlevirus were collected and ground finely in a food blender in 2 volumes of 0. 5 mol citrate buffercontaining 5% sucrose, 0. 01 mol EDTA, 1% PVP and 0.1% thioglycollic acid. Filtrateyielded through nyloncloth was clarified with 20% chlorform and 8% n-butanol. Aqueousphase was treated with 2 cycles of adding 8% PEG6000 and 3% NaCI,combined with highspeed centrifugation, to separate virus from plant materials. As a rseult, partially purifiedvirus preparation was obtained after resuspension of pellets in 0. 01 mol borate buffer. TheA260～ 290/280 absorption ratio of the preparation is 1. 12, the Amax is at 258nm, Amin at244nm, virus concentration is 2. 75 mg/ml. By four intramuscular injections given to a rabbit, the specific antiserum with titre 1: 1 024 in microprecipitin test was produced. Becauseof small amount of plant protein in the purified virus preparation, care must be taken toavoid non-specific reaction confusing index results when using the present antiserum in virusdetection.
