摘  要:用黄瓜花叶病毒接种矮牵牛,取发病植株的叶片、花芽和茎尖进行培养,用间接ELISA和离体蚕豆叶片进行检测,结果表明:用叶片、花芽以及大于0.3~0.5mm的茎尖培养,不能获得无毒苗,从病株上剥取0.15~0.25mm的茎尖培养可获得6.6%的无毒苗;连续高温处理病株8d结合茎尖(0.3~0.5mm)培养,可获10%的无毒苗;分别变温处理(38℃8h+22℃16h)16d和30d结合茎尖(0.3~0.smm)培养,均可获得11.1%的无毒苗。The leaves, flower buds and meristem-tips from the infected petunia plants inoculated with CMV were cultured and the virus in calli and plantlets was checked by indirect ELISA and by inoculation on detached broad-beau leaves. The results showed as follows: (1)virus-free plantlets were not obtained as the leaves, flower-buds and meristem-tips (more than 0. 3mm) were cultured; (2) 6. 6% plantlets were apparently free from CMV as 0. 15~0. 25mm meristem-tips were cultured; (3) 10% virus-free plantlets were produced from the cultures of meristem-tips(0. 3-0. 5mm) from the infected plants that had been continuously treated at 40℃ for & days; (4)the same percent (11. 1%) of virus-free plantlets were obtained as the meristem-tips (0. 3~0. 5mm)were cultured from the infected plants that had been treated with alternating temperature in diurnal circles 38℃ 8h+22℃ 16 h for 16 or 30 days, respectively.
