摘  要:本实验应用原位杂交组织化学技术对GABAA受体的α6亚单位的mRNA在大鼠脑内的分布状况进行了观察。除得到与以往的报道相同的结果即发现小脑浦肯野氏细胞含α6亚单位mRNA外,还发现此mRNA也分布于耳蜗核。在背侧耳蜗核,标记神经元主要见于颗粒细胞层和中央区内;而在腹侧耳蜗核,标记神经元只见于颗粒细胞层。这些结果表明α6亚单位是耳蜗核中某些神经元的GABAA受体的重要组成成分。另外,在耳蜗核旁胶质成分区中也见到一些标记细胞,其意义有待进一步研究。Using in situ hybridization histochemistry with synthesized oligonucleotide probe, we investigated the distribution of neurons containing α6 subunit mRNA of GABAA receptor in the rat brain. Fresh frozen sections were used in the experiment.To do in situ hybridization , the sections were prehybridized with 4×SSC containing 1×Denhardt's solution and dehydrated through a graded ethanol series (70%~100%). Hybridization were performed by incubating sections with the buffer containing [α-35S] dATP-labeled probes at 41℃. After hybridization. the sections were rinsed 3 times in 1×SSC at 55℃for 20 minutes each time. Then the sections were dehydrated through a graded ethanol series (70%~100%) and coated with Kodak NTB-2 emulsion. After exposure for 4~6 weeks at 4 ℃, the sections were developed in D-19 (Kodak) developer,fixed with photographic fixer, and washed with tap water. The result showed that labeled neurons were only seen in the granule cell layer of cerebellum and the cochlear nucleus. In the dorsal cochlear nucleus , the labeled neurons were located in the granule cell and central regions. while those in the ventral cochlear nucleus were found only in the granule region. In addition , some labeled neurons were also detected in the paracochlear glial substance. These results indicated that α6 subunit is a very important component of the GABAA receptor in the cerebellum and cochlear nucleus.
