摘  要:用免疫电镜技术观察了豚鼠心脏房间隔内CGRP-IR神经纤维或终末。结果显示该部位存在CGRP-IR阳性和阴性的神经纤维和终末。阳性者伴随心肌细胞、血管或一种囊泡状突起而行,其内均含多量小颗粒囊泡,少量大颗粒囊泡和少许小无颗粒囊泡及个别线粒体。这些纤维与终末的特点与含去甲肾上腺素(NA)的纤维或终末相似,提示豚鼠心脏中存在CGRP与NA共存的纤维或终末。这种结果与心脏神经中CGRP与NA关系密切的现象相吻合。阴性者则不具备上述特征。The present findings provide evidence that Calcitonin Gene-related Peptide (CGRP) can induce positive inotropic and chronotropic response of human heart and cause vasodilatation of human coronary arteries. CGRPimmunoreactivity is widely distributed in mammalian heart nerve tissues. The majority of CGRP-immunoreactive (CGRP-l) nerve fibers are presumed to be sensory in origin. The others is suggested to be NANC (nonadrenergic noncholinergic) nerve fibers.In the present study, we examined the CGRP-l nerve fibers and terminals in the in the guinea pig interatrial septum (IAS) at the ultrastructural level to identify the character of them. Tissues were obtained from guinea pigs (300~500g.body weight, n=3) which were perfused with 200 ml normal saline followed by 400 ml of a solution containing 4% paraformaldehyde and 0. 05% glutaraldehyde in 0.1 mol/L sodium phosphate buffer (PB,pH 7. 2). IAS were removed and immersed in fresh fixative for 2h. The tissues were then left in 20% sucrose in 0. 1 mol/L PB for 2h. And 50 μm sections were prepared with vibratome. A peroxidase-anti-peroxidase (PAP) technique was used, in which rabbit polyclonal antibody to CGRP (Amersham, America) in dilution 1: 1000 and goat-anti-rabbit IgG (Shanghai Medical University) 1:100 and PAP complex (same source as above) 1:200 were used. The positive areas were then selected and processed for electron microscopy. The control was done with rabbit serum, diluted in the same concentration as, and used instead of, CGRP antiStrongly CGRP-l fibers and terminals (0. 4~1. 5μm in diameter) were visualised in the guinea pig LAS. They were surrounded by a thin fibrillar basement membrane and were not invested with a Schwann cell sheath. These fibers or terminals traveled along the myocardium (80~250 nm in distance), or vascular smooth muscles (70 nm in distance), or a vascular structure (500~1000nm) in distance) onthe surface of the cardiac muscle cells.
