摘  要:本实验采用热休克蛋白70(HSP70)核酸分子杂交方法,检测了自发性高血压大鼠(SHR)和正常血压大鼠(WKY)离体培养的主动脉平滑肌细胞(ASMC)受热刺激后HSP70mRNA水平的变化以及整体动物肝组织HSP70mRNA的水平,并对肝组织基因组DNA进行了限制性酶切片段长度多态性(RFLP)分析。结果表明:37℃培养的SHRASMC及整体SHR肝组织HSP70mRNA的基础表达水平均低于WKY鼠,SHRASMC受热刺激(42℃15min)后2h,HSP70mRNA表达增加的程度明显大于WKY鼠。肝组织基因组DNA经BamHI酶切后,SHRHSP70基因缺失一条约5.6kb的片段。本结果提示SHRASMC对热敏感,HSP70基因异常可能与高血压发生有关。The level of heat shock protein 70 messenger RNA was examined in aorticsmooth muscle cells(ASMC)and hepatic tissue of Wistar-Kyoto(WKY)rats andspontaneously hypertensive rats(SHR).The restriction fragment length polymorphism(RFLP)was also analysed in hepatic DNA.It was demonstrated that the basiclevel of heat shock protein 70messenger RNA in ASMC and heretic tissue of SHRwere lower than that of the WKY rats.The extent of heat shock Plotein 70 messengerRNA accumulated in ASMC was higher in SHR than in WKY rats when the cells wereheated by immersing the culture bottles in 42℃ water bath for 15 min and allowed torecover at 37℃ for 2 h. In contrast with WKY rats,the Bam HI restricted bepeticDNA showed that a fragment about 5. 6 kb was lost in heat shock protein 70 genefrom hepatic DNA of SHR.The results suggest that the ASMC of SHR is more sensitive to heat stress than that of WKY rats and the changes of heat shock protein gene sequence may be related to genetic hypertension.
