摘  要:用显微注射法导入外源SMTPGH基因的早期兔胚进行体外培养,并用PCR技术对单个胚中的外源基因进行了检测,以探讨SMTPGH基因在早期兔胚中的滞留情况。研究结果表明早期转基因兔胚在Tc199+10%FCS培养液中有75%发育到囊胚期;其外源基因在8细胞期前没有丢失,随后有逐渐丢失的现象。到胚胎的发育后期,其外源基因的滞留率接近于整合率。We studied -the culture of early transgenic rabbit embryos containing the SMTPGH gene with different medium. The retention of foreign gene in the embryo was detected with PCR and nonradioactive labeling method. The early transgenic rabbit embryos can develop into blastocyst stages with 75% of developmental rate in the cultural medium of Tcl99+10%FCS. There were not any apparent changes found before 8-cell stage. After 8-cell stage, the foreign gene either was integrated on chromosomes or was lost gradually. But finally, the retention rate of foreign gene should be equal to the integration rate of the gene.
