When the whole peptide chain has been assembled, the peptide-resin was treated with HF containing 10% p-cresol at 0℃for 1 hour. After the removal of HF and p-eresol, free peptides were extracted with 1N acetic acid and lyophilized. GIP was purified by gel filtration on TSK column and ion-exchange chromatography on DEAE-Sepharose OL-6 B column.The sgnthetie GIP was identified by PAGE, electrofocusing, amino acids composition, HPLOC and checked in chcmical structure by sequence determination. The in vivo activity of GIP to stimulate insulin release and its antigenicity were also reported in this article.
