摘  要:以感染丛枝病原(MLO)的泡桐组培苗为试材,大量切取长度在0.3mm~0.5mm范围内的微茎尖,在MS和附加0.1mg/LIBA和1~7mg/L6-BA的培养基上进行脱毒试验。根据外植体培养过程的病状有无.病源MLO的DAPI荧光显微镜和电子显微镜检查结果,证明微茎炎培养能够获得脱除泡桐丛枝病原MLO的试管苗,其脱毒率在14.29~28.57%之间。脱毒的两个无性系试管苗经连续继代培养一年以上,其生长性状正常且稳定。试验还显示了微茎炎培养脱毒率受采用的茎尖大小、切割技术、培养基中的激素比例及无性系的不同等因素的影响较大。The experiments on eradication of paulownia witches f broom agentmycoplasmalike organism (PWB-MLO) by means of meristem tip culture were conducted. The test results show that culture of 0. 3 ̄0. 5 mm meristem tip was effective in producing about 14. 29 ̄ 28. 57 % MLO-free plants from paulownia plantlets infected with PWB-MLO in the modified MS medium supplemented with 0. 1 mg/L IBA and 2. 0 mg/L 6-BA. The screening as well as further identification of MLO-free plantlets were achieved by the combination of the symptom observation, DAPI fluoreseence microscope and electrom nicroscope detection of plantlets before and after their subcultures. The acquired disease-free plantlets of two clones, C85-028 and C443, have been subcultured on MS or modified MS media lashing for one yeal or more, and always preserve stable growth character possessed by the healthy plant. It is also found that the rate of ILO-free plantlet obtained by meristem tip culture was affected, at differentdegrees, by the size and excising skill of metistem tip, the concentration and proportion of growth regulators in the media as wen as the variation of the clones.
