摘  要:木文对高分子共聚物PLGA(9:1)和PLGA(8:2)包制的18-甲基炔诺酮(LNG)微球长效避孕注射剂进行了研究。结果显示二种注射剂具有比LNG微晶释药缓慢、平稳的性能,体内生物半衰期(t1/2)分别为18.14和16.72天,LNG微晶混悬液仅为6.01天(P<0.01);抑制大鼠性周期的天数分别为41.30和39.60天,也比LNG微晶(24.90天)时间长(P<0.01)。微球中载体PLGA(8:2)体内降解速度比PLGA(9:1)快,证实调节丙交酯(LA)与乙交酯(GA)的配比可以控制载体的降解时间。This paper describes that the release rate,the biodegradation of polymer and the estrus suppression of two kinds of polymer microspheres containing LNG in vivo.The release test in vivo was carried out in female rats by intramuscular injection of microspheres and the serum LNG levels were analyzed by RIA. The effect of estrus suppression in rats were observed with vaginal smears.The results showed that the rate of LNG releaso from microspheres was slower than that from microcrystal of LNG.With the increase of LA proportion of ALGA, the LNG release rate of microspheres decreased.The biodegradation test of polymer indicated that PLGA(8:2)degraded much faster than PLGA(9:1)The degradation rate was significantly enhanced with the inerease of glycolic acid(GA)contend in copolymers.
