摘 要:用正交试验确立了超声提取蛇足石杉生物碱的最佳条件。以石杉碱甲和石杉碱乙回收率为指标 ,考察了溶剂倍数、溶剂浓度、超声时间、超声功率等因素的影响。结果表明 ,在室温下超声提取的最优条件为 :酸浓度 0 . 8% (v/v) ,液固比例 2 0 :1,超声功率 6 0 0W ,超声 15min。三次重复实验所得石杉碱甲和石杉碱乙回收率分别是 97. 3%和 93. 5 % ,相对标准偏差分别为 1. 31%和 1. 4 0 % (n =3)。与传统的回流提取工艺相比 ,过程时间从2h缩短为 15min ,回收率提高了 10 %以上。Extraction of huperzine A and huperzine B from Huperzia serrata was enhanced with application of ultrasonic field in the aqueous solution containing H 2SO 4.An orthogonal design method was used to search for the best experimental condition.The experiment was carried out at room temperature with variation of ultrasonic power,processing time,acid concentration and ratio of the solution to the plant solid.The best result was obtained when the ultrasonic power was 600 W,processing time 15 min,H 2SO 4 concentration 0.8%,and the ratio of the solution to the plant solid 20:1.Under the optimized condition,the average recoveries of huperzine A and huperzine B were 97.3% and 93.5% respectively.The relative standard deviations for three repeated experiments were 1.31% and 1.40%(n=3).Compared with the traditional method of reflux extraction,the novel extraction method reduced the process time from 2 h down to 15 min with 10% increase in the recovery.
