摘 要:以根癌农杆菌(Agrobactriumtumefaciens)LBA4404菌株介导异戊烯基转移酶基因(ipt)转化切花菊(Dendrathemagrandiflorum),对转化材料进行连续的Kan抗性筛选,获得69株抗性株。部分植株经PCR和Southernblot检测呈阳性,对照均为阴性;ELISA检测抗性株衰老同期叶片内源iPAs含量是对照株的4.7倍。田间观察株型、叶色、花芽分化等性状和对照相比出现差异。叶绿素含量测定显示,抗性株叶片在花芽分化期、盛花期和衰老期总叶绿素含量分别是对照株的1.47、2.26和2.60倍。Transgenic cut chrysanthemum(Dendrathema grandiflorum cv. Huang Xiufang) plants were mediated by Agrobacterium tumefaciens LBA4404 and 69 transformants were obtained after successive Kan resistance selection. Some of the Kan-resistant plants were detected to be positive by PCR and Southern blot. During the aging phase in the field, the content of endogenous iPAs in the transgenic leaves was 4.7 times higher than that in CK by ELISA. The model of plant, the leaf color and the flower bud differentiation were different from the control. The chlorophyll contents were genetic tranformation 1.47-, 2.26-, 2.60-fold of those in the controls at the flower bud differentiation, the blooming florescence and the senescence phases respectively.
