摘 要:以控制BooroolaMerino绵羊高繁殖力的骨形态发生蛋白受体IB(bonemorphogeneticproteinreceptorIB,BMPR-IB)基因为候选基因,采用PCR-SSCP技术检测BMPR-IB基因在高繁殖力绵羊品种(小尾寒羊、湖羊)以及低繁殖力绵羊品种(多赛特羊、萨福克羊)中的单核苷酸多态性,同时研究该基因对小尾寒羊高繁殖力的影响。结果表明:高繁殖力的小尾寒羊和湖羊在BMPR-IB基因编码序列第746位碱基处发生了与BooroolaMerino绵羊相同的突变(A746G),而低繁殖力的多赛特和萨福克绵羊则没有发生这种突变。小尾寒羊B等位基因频率为0.716,+等位基因频率为0.284;湖羊B等位基因频率为0.941,+等位基因频率为0.059。高繁殖力绵羊品种(小尾寒羊、湖羊)与低繁殖力绵羊品种(多赛特羊、萨福克羊)之间BMPR-IB基因型分布差异极显著(P<0.001)。BB基因型小尾寒羊第一胎和第二胎产羔数分别比++基因型多产0.92只(P<0.01)和1.02只(P<0.01)。BMPR-IB基因型在几个绵羊品种之间的分布以及各种基因型绵羊的实际产羔两方面的研究结果都证明BMPR-IB基因是影响小尾寒羊和湖羊高繁殖力的一个主效基因,可以用于对绵羊产羔数的辅助选择。Bone morphogenetic protein receptor IB (BMPR-IB) gene which controls fecundity of Booroola Merino ewes was studied as a candidate gene on the prolificacy of Small Tail Han sheep. Single nucleotide polymorphism of BMPR-IB gene was detected in both high fecundity sheep breeds (Small Tail Han sheep and Hu sheep) and low fecundity sheep breeds (Suffolk sheep and Dorset sheep) by PCR-SSCP. The results indicated that there was a same mutation (A746G) of BMPR-IB gene in both Small Tail Han sheep and Hu sheep as that in Booroola Merino ewes. That mutation did not exist in both Suffolk sheep and Dorset sheep. In Small Tail Han sheep, frequencies of B allele and + allele were 0.716 and 0.284 respectively. In Hu sheep, these frequencies were 0.941 and 0.059. The BMPR-IB genotype distributions were high significantly different (P < 0.001) among high fecundity sheep breeds (Small Tail Han sheep and Hu sheep) and low fecundity sheep breeds (Suffolk sheep and Dorset sheep). The ewes with genotype BB had 0.92 (P < 0.01) and 1.02 (P < 0.01) lambs more than those with genotype ++ in the first parity and the second parity in Small Tail Han sheep, respectively. These results intensively demonstrated that BMPR-IB gene is a major gene that affects the prolificacy in both Small Tail Han sheep and Hu sheep, and could be used as a molecular genetic marker to select the litter size in sheep.
