摘  要:钙火花(Ca2+sparks)现象是近年来在骨骼肌、心肌兴奋收缩偶联研究中发现的重要生理现象,但目前确切的机制还不清楚。我们利用激光扫描共聚焦显微镜对经Flou 3AM染色的人工剥离的蟾蜍骨骼肌肌纤维进行连续扫描分析,在骨骼肌肌纤维中捕获到钙火花现象,并对钙火花现象的发生进行了初步探讨。Fluorescence probe Fluo-3AM can be luminary in confocal microscope after combining Ca^(2+), so we can measure and test nomadic Ca^(2+) in cytoplasm by this way. Some confocal laser scan images were obtained after calcium in muscle fibers signed with Fluo-3AM. A group of luminescent substance emerged close to sarcolemma intracellulare after electron stimulation, and the volume enlarged and the brightness augmented with the time passing. At the 64s, the volume reached maximum. That is the Ca^(2+)sparks changing with time in skeletal muscle.The generation of Ca^(2+) sparks are necessarily related with the calcium release from SR, so we need to confirm it further by calcium determination and physiological experiments, which may be a theoretical support to a series neural muscular pathological changes.
