摘 要:为检测和证实主动外排系统 (外输泵 )在细菌多重耐药机制中的作用 ,用四环素和水杨酸钠对大肠杆菌质控株进行了体外人工诱导 ,筛选出多重耐药株 ,再用能量抑制剂 CCCP(Carbongl cyanide m- chloropheyhydrazone)对耐药株和质控株进行了环丙沙星摄取抑制试验。结果 :在耐药株 ,其菌体内环丙沙星稳态浓度明显低于质控株 ;多重耐药株在有 CCCP存在时 ,菌体内环丙沙星的浓度随时间的延长而递增 ,而无 CCCP的抑制时 ,菌体内环丙沙星的浓度递增缓慢 ;在质控株 ,不论有无 CCCP存在 ,菌体内环丙沙星的浓度都随时间的延长而呈递增趋势 ,且保持较耐药株明显高的水平。结论 :体外诱导多重耐药大肠杆菌多重耐药性的产生 。The objective of the experiment is to detect and confirm the effect of active efflux system in the mechanism of bacterial multi-drug resistance.In vitro,tetracycline and sodium salicylate was used to induce the control E.coli strain artificially,and at the same-time,multiple resistant strains were screened.Finally,estimated the action of CCCP(Carbongl cyanide m-chloropheyhydrazone) on absorption of ciprofloxacin in the resistant strains and control strains.Results:The stable concentration of ciprofloxacin is obviously lower in the resistant strains than in the control strains.Presence of CCCP made the concentration of ciprofloxac in increased progressively by time,without the action of CCCP,the concentration of ciprofloxacin increased slowly.In the control,the concentration of ciprofloxacin increased progressively by time and maintained much higher level than that of resistant strains no matter the presence or abcence of CCCP.Conclusion:It is substantiated that the multi-drug resistance induced in vitro should be mediated by the active efflux system.
