摘 要:应用自组装毛细管电泳-安培检测装置,建立了同时分离检测抗精神病药奋乃静与氟奋乃静的新方法.在1050 mV(vs. Ag/AgCl)检测电位下,奋乃静与氟奋乃静的检出限分别达0.060 mg/L与0.075 mg/L.在最佳检测条件下,两组分的响应电流与浓度之间有着良好的线性关系,线性范围为0.75~75 mg/L.将方法用于药物片剂以及模拟尿样和血清样品中奋乃静和氟奋乃静的测定,结果令人满意.A sensitive method based on capillary electrophoresis ( CE) with amperometric detection (AD) has been developed for the separation and determination of perphenazine and fluphenazine. The working electrode used in here was a 300 mu m diameter carbon disk electrode (vs. Ag/AgCl) operated in a wall-jet configuration. The optimum conditions were obtained with a working potential of 1050 mV and a 75 cm capillary (360 mm, 25 mu m i. d.) at 18 kV in the electrophoretic medium of 20 mmol/L phosphate buffer solution ( pH = 2. 7). Under the optimum conditions, the current response was linear over about two orders of magnitude with detection limits (SIN = 3) of 0. 060 mg/L for perphenazine and 0. 075 mg/L for fluphenazine, respectively. The proposed method has been successfully applied to determine the content of perphenazine and fluphenazine in perphenazine tablet, fluphenazine tablet, spiked urine and rabbit serum samples with satisfactory results.
