摘 要:目的 探索血管紧张素转换酶(angiotensionconvertingenzyme,ACE)基因多态性与冠心病的关联分析。方法 应用聚合酶链反应(PolymeraseChainReaction,PCR)技术对 100例住院冠心病患者及 90例正常对照者进行ACE基因多态性测定,并对冠心病患者进行A型行为类型问卷(TypeABehaviorPatternScale,TABP)测定。结果 冠心病组的DD基因型频率 (34% )及D等位基因频率 (66% )均显著高于正常对照组(14. 4%, 44. 4% ),且RRDD=3. 05,RRD=2. 43,P<0. 05,表明冠心病与ACE基因的DD基因型、D等位基因呈显著正关联;具有A型行为的冠心病患者的DD基因型频率及D等位基因频率均显著高于非A型患者(P<0. 05);携带DD基因型的冠心病患者的A型行为问卷的TH因子 (15. 88±3. 16)、CH因子分(14. 88±2. 89)极显著地高于携带ID及Ⅱ基因型患者的因子分 ( 12. 89±4. 53, 12. 52±3. 88, 13. 00±1. 41, 10. 50±0. 71) (P<0. 01)。结论 ACE基因多态性可能是冠心病发生发展过程中重要的危险因素之一,其DD型和D等位基因与冠心病患者的A型行为有一定的联系。ObjectiveTo explore the relation between angiotensin converting enzyme (ACE) gene polymorphism and coronary heart disease (CHD). MethodsThe Polymerase Chain Reaction (PCR) technique were applied to compare the polymorphism of ACE gene between 100 CHD patients and 90 normal control. Type A Behavior Pattern Scale (TABP) were tested with CHD patients. ResultsThe frequency of DD genotype(34%) and rare D allele(66%) of the CHD patients group was significantly higher than that of controls(14.4%,44.4%), and RR DD=3.05,RR D=2.43,P<0.05,showed that DD genotype and rare D allele of ACE gene were significantly positive correlated with CHD. The CHD patients with Type A behavior had significantly higher frequency of DD genotype and rare D allele than patients with Type B. The CHD patients with DD genotype had significantly higher TH(15.88±3.16) and CH(14.88±2.89) factor score than patients with ID and Ⅱ genotype(12.89 ±4.53,12.52±3.88;13.00±1.41,10.50±0.71). ConclusionsThe polymorphism of ACE gene may be one of the important factors for CHD, and the DD genotype and rare D allele of ACE gene in the CHD patients had some relationship with Type A behavior.
