摘 要:目的建立人血中精神药物的GC-MS测定方法。方法样品在pH12条件下用乙醚萃取,SKF_525A为内标。以DB-5MS石英毛细管柱、EI源、不分流进样测定人血中四种精神药物。结果人血中四种精神药物与内标物分离良好,血药浓度在0.2-1.0μg,/ml范围内呈线性关系,R2≥0.9728,最低检出限为0.01μg(S/N≥3),血中精神药物的最低检出浓度为0.05μg/ml(S/N≥3),提取率72.5%~89.3%,RSD≤5.17%。四种精神药物日内差RSD≤3.7%(n=5)。结论本法可用于精神药物的血药浓度监测。Objective To develop a method for the determination chlorpromazine, clozapine, benzhexol and penfluridol in human blood by GC-MS. Methods The specimens were alkalized and extracted with ether. The DB-5MS quartz capillary column (0.25mm×30m×0.25μm) with Electron Impact Ion source (EI) were used for determination four antipsychotic drugs with SKF525A as the internal standard. Results As established in the method, four antipsychotic drugs were well separated from the internal standard. The linear range was from 0.2μg/ml to 1.0μg/ml (R2≥0.9728). The limit of detection was 0.01μg (S/N≥3), and extraction efficiencies ranged between 72.5% and 89.3% with RSD≤5.17%. The intra-precisions of assay for four antipsychotic drugs were less than 3.7% (n=5) in human blood. Conclusion The GC-MS method developed is sensitive, accurate and reliable for determination of the antipsychotic drug concentration in human blood.
