草木樨状黄芪单细胞培养再生植株及其影响因素的研究  被引量:4

摘  要:从草木樨状黄芪的子叶和胚轴诱导的愈伤组织中选取结构松脆的颗粒状愈伤组织,用液体培养基S_(12)进行悬浮培养建立了悬浮细胞系。取换液3天左右生长旺盛的悬浮系材料分离单细胞,用基本培养基S_(12)与不同浓度的植物血球凝集素组成的系列培养基MC_1-MC_6进行液体浅层静置暗培养;形成小愈伤组织后,经M_3(MS+2,4-D2mg/L,NAA0.2mg/L,KT1mg/L)培养基增殖,MR_2培养基诱导分化出苗,再经RI(1/2MS+IBA0.8-1.5mg/L)培养基诱导生根,获得完整再生植株。研究表明适量的植物血球凝集素对草木樨状黄芪的单细胞培养是有益的。本文还对影响悬浮系细胞和单细胞培养的其他一些因素也进行了探讨。The calli with a higher capacity of redifferentiation selected from somaclonal calli of As-tragalus melilotoides Pall.were transfered to fluid medium S_(12) ,which had been screened frommany different media and was suitable for suspension culture of this plant,and the cell-sus-pension line was obtained.A great number of monocells were seperated from the cell-sus-pension by the filtration and then cultured in liquid media MC_1-MC_5.After minicalli wereformed from the menocells,they were transfered into the differentiation medium.Youngshoots were regenerated from the monocell calli.After the roots were induced on medium RI(1/2 MS+IBA 0.8-1.5mg/L)the shoots developed into whole plants. It was shwon thatPHA promoted the cell division and development in monocell culture. The other factors af-fected the monocell cultures were also discussed.
