炔诺酮肟对假孕家兔卵巢黄体的组织学、组织化学及超微结构变化的观察

摘  要:家兔从假孕当天按每天2mg/kg或4mg/kg灌服炔诺酮肟,共3天,观察卵巢黄体组织学、组织化学及超微结构的改变。结果表明,给药两组家兔黄体细胞空泡变性,3β-羟基甾体脱氢酶(3β-HSD)活性减弱。2mg/kg剂量组黄体细胞高碘酸-Schiff氏PAS反应(PAS)、碱性磷酸酶(AKP)、酸性磷酸酶(ACP)及琥珀酸脱氢酶(SDH)活性无变化。4mg/kg剂量组AKP活性减弱,ACP活性增强,黄体细胞胞浆内光面内质网减少,脂滴大小不等,脂滴聚集及排空等变化。并讨论了药物引起黄体细胞形态学变化与卵巢孕酮生物合成受阻的关系。Rabbits were given norethisterone oximeorally at a daily dose of 2-4 mg /kg from the first day of pseudopregnancy for 3 days. Histological, histochemical and ultrastructural changes of lutea cells were observed. It was found that norethisterone oxime induced vacuolar degeneration of lutea cells, and the activity of 3β-hydroxy-steroid dehydrogenase decreased. PAS reaction of lutea cells and the activity of ACP, AKP.and SDH remained unaltered in treatment group(2 mg/kg.day), but the activity of AKP decreased, while that of ACP increased in treatment group (4 mg/kg.day). Electron microscopic examination indicated that in the treatment group(4 mg/kg.day) smooth endoplasmic reticula in lutea cells decreased and lipid droplets of various sizes were noted with the sign of exhaustion and conglomeration. The relation between morphological changes of lutea cells and the biosynthesis of progesterone is also discussed.
