摘 要:目的 探讨硼对过量氟所致实验大鼠骨与软骨损害的治疗作用。方法 46只 SD大鼠 ,4~ 5周龄 ,体重 5 0~ 90 g,随机分为 3组 :对照组 (饮蒸馏水 )、低过剂量氟组 (饮用含氟化钠 1 1 0 .5mg/L蒸馏水 ,F-5 0 mg/L ) ,高过剂量氟组 (饮用含氟化钠 2 2 1 mg/L蒸馏水 ,F-1 0 0 mg/L)。实验后 3个月 ,过量氟大鼠模型成功。造模后用硼治疗 ,各过剂量氟组大鼠又分为过剂量氟组与过剂量氟加硼治疗组 ,各治疗组大鼠饮水含氟量与摄氟组相同 ,同时摄取补硼饮食。治疗两个月后观察实验大鼠血清游离氟和硼含量、四肢骨氟含量、血清 NO含量 ,股骨生物力学、骨密度及大鼠肋骨、肋软骨交界处的病理形态学变化 ,肋软骨 型胶原表型表达和蛋白多糖变化。结果 1 )硼治疗后大鼠血清和四肢骨中游离氟及血清 NO含量减少 ;2 )光镜下肋骨生长板的损伤显著恢复 ;3)和对照组及摄氟组相比 ,治疗组大鼠蛋白多糖和 型胶原表达恢复。结论 硼对氟中毒导致自由基代谢异常及软骨基质中蛋白多糖和 型胶原的异常表达有改善作用。Objective To understand the therapeutic effect of boron on damages of bone and cartilage in rats in-gested excessive fluoride.Methods46Sprage-Dawley rats after the birth of4-6weeks were randomly divided into five groups:the control(C,intake the distilled water),the low excessive fluoride dose group(LF,intake the 50ppm F - in distilled water),the high excessive fluoride dose group(HF,intake the100ppm F - in distilled water),the therapeutic groups with the supplemental boron dietary for LF(LFB:intake the50ppm F - in distilled water with the supplemental boron dietary,and the therapeutic groups with the supplemental boron dietary for HF (HFB:intake100ppm F - in distilled water with the supplemental boron dietary).Contents of free fluorine,boron and Nitric oxide(No)in serum,fluorine in bone,bone biomechanics of femur and BMD were assayed at the end of 2months from the beginning of boron therapy.The morphological changes,proteoglycan stained by toluidine blue and the expression of typeⅡcollagen in rib growth plate cartilage(GPC)by immunohistochemistry were observed in all of groups.Results(1)The free fluorine contents in serum and bone were decreased,and the NO level de-creased,respectively.(2)The damages of rib GPC under optical microscope were recovered significantly.Proteo-glycan and the expression of typeⅡcollagen in rib cartilage of rats in groups of HFB and LFB were recovered con-trast to the control and groups of LF and HF.Conclusion Boron has some therapeutic effects against fluoride toxic-ity and improve the pathological changes in experimental rats with skeletal fluorosis.
