摘 要:目的:探讨奥曲肽在体外对人胆囊癌细胞VEGF表达的影响。方法:体外培养人胆囊癌细胞株GBC-SD,用RT-PCR法检测VEGFmRNA水平,用ELISA法检测培养液上清中的VEGF蛋白水平。结果:GBC-SD高水平表达VEGF;经奥曲肽作用后,VEGFmRNA/β-actin相对系数显著下降,呈剂量依赖效应;奥曲肽也显著降低培养上清液中的VEGF浓度,抑制作用随时间延长和奥曲肽剂量增加而增大。结论:奥曲肽能下调GBC-SD的VEGF的mRNA和蛋白表达水平。Objective: To investigate whether octreotide can inhibit the vascular endothelial growth factor (VEGF) expression of human gallbladder cancer cells in vitro. Methods: A human gallbladder cancer cell line, GBC-SD, was cultured in vitro. VEGF mRNA was measured quantitatively by RT-PCR method, and VEGF protein concentration in the culture supernatant by enzyme linked immunoabsorbent assay. Results: Both VEGF mRNA and protein were detected in high levels in GBC-SD cells. In 36 hours the concentration of VEGF continuously increased. Octreotide significantly decreased the expression of VEGF mRNA and protein, with dose and time-related effects. Conclusion: Octreotide downregulates the expression of VEGF in gallbladder cancer cells in vitro.
