抑前胸腺肽溶液构象的2DNMR初步分析

摘  要:抑前胸腺肽PTSP是一种重要的新发现的昆虫神经肽,能够在体内和体外条件下抑制促前胸腺素PTTH对蜕皮激素合成的促进作用.实验材料PTSP以固相方法合成.用2 D NMR技术来考察PTSP的DMSO- d6 溶液,并记录COSY,NOESY和1 3C HMQC- COSY谱用于构象分析.利用2 D COSY和NOESY归属了1 H NMR谱的全部共振峰;再利用HMQC- COSY使全部αC和βC共振峰也得到了归属.分析得到肽链的1 H和1 3C的化学位移和质子间的NOE,结果表明PTSP在DMSO- d6 中构象最有可能为无规卷曲.此构象可能是PTSP与其受体结合前,邻近前胸腺腺体细胞膜表面时的一种状态。Bombyx mori prothoracicostatic peptide (Bom-PTSP) is an important insect neuropeptide, which inhibits the stimulation of prothoracicotropic hormone on ecdysteroidogenesis by prothoracic glands in vivo and in vitro. In the present paper, PTSP was obtained by so1lid phase method. Its solution in DMSO-d 6 was examined by 2D NMR technique. Then the COSY, NOESY and 13 C HMQC-COSY spectra were applied for the analysis of the conformation. All resonance assignments of 1H NMR were made by using two-dimensional correlation spectroscopy. The problem of sequential resonance assignments was solved by the analysis of NOESY spectroscopy. The chemical shifts of 1H and NOE suggest that PTSP exists mainly as random coil in DMSO-d 6.
