摘  要:目的:研究中药泽泻提取物中的3种化学成分在体外对尿草酸钙结石形成的抑制作用,筛选泽泻抑制尿草酸钙结石形成的活性成分。方法:采用现代植化和生物活性导向分离的方法分离提取泽泻的3种化学成分,并应用标准种晶技术分别在不同时间(1,2,3h)评价不同浓度(100,500,1000mg·mL^(-1))的3种化学成分对草酸钙结晶生长的抑制作用。结果:化学成分Ⅰ对草酸钙晶体生长抑制作用最强,抑制指数最高达89.43%,且该抑制作用在一定范围内呈时间、剂量依赖性。其结构经初步鉴定主要为四环三萜类化合物。结论:泽泻的化学成分Ⅰ在体外对草酸钙晶体的生长有较好的抑制作用,可能是泽泻抑制尿草酸钙结石形成的活性成分。Objective: To investigate the inhibitory effect of the three active constituents from Alisma orientalis on the formation of urinary calcium oxalate calculus in vitro. Methods: A unique bioactive-targeted extraction technique was used to isolate three constituents from Alisma orientalis. The inhibitory effects of the three constituents whose concentrations were at 100, 500 and 1000mg· mL^(-1), respectively, were evaluated in vitro using a standard seeded crystallization technique at 1, 2 and 3 hours post crystallization, respectively. Results: The constituent Ⅰ had the strongest inhibition with the inhibitory index of 89.43%, and showed the inhibitory dependency of the crystallization period and concentration in a certain range. The structure of the constituent Ⅰ was elucidated as tetracyclic triterpenoid. Conclusion: The constituent Ⅰ in Alisma orientalis played a key role in the inhibition of the formation of calcium oxalate calculus.
