摘  要:研究了荧光动力学分析法检测超氧自由基抑制剂抗氧活性的新方法.在 Tris HCl缓冲溶液中(pH=8.20),邻苯三酚自氧化产生超氧自由基和有荧光效率的自氧化产物.在433.0 nm激发波长和494.5 nm发射波长条件下,该自氧化产物时间扫描曲线的斜率与超氧自由基抑制剂的浓度有相关性.利用抗坏血酸、槲皮素、芦丁 3 种常见抗氧化剂对新方法进行了验证并将新方法与单扫描极谱法(SSP)、紫外分光光度法(UV)作了比较.结果表明新方法测活灵敏度优于UV法,而与SSP法相当,但比SSP法操作简单、准确度好.A new method of determination antioxidant activity of scavening on superoxide radicals has been studied by Fluorescent Kinetic Analysis(FKA). The superoxide radicals and autoxidation products with fluorescence efficiency were produced by autoxidation of pyrogallol in the Tris-HCl buffer solution (pH 8.2). The slope of time scan curve of the autoxidation products were related to the concentration of scavening in Ex=433.0 nm and Em=494.5nm. The method has been proved by Ascorbic Acid, Quercetin and Rutin, and were comparaed with Single Sweep-polarography(SSP) and scripture Ultraviolt and Visible Spectrophotometry(UV). Results showed the sensibility of FKA were higher than scripture UV, and basically equivalent with SSP, but FKA were more simple and accurate.
