摘 要:目的: 研究mCcd1新基因在小鼠胚胎发育过程中的基因表达,为其功能研究提供线索. 方法: 用地高辛标记mCcd1 RNA探针, 对E9~E11.5小鼠整体胚胎进行原位杂交,检测胚胎发育过程中基因的表达.结果: 在E9~E11.5都有不同程度的表达,E9和E10.5全身都表达,但表达量低.在E11.5的小鼠胚胎开始神经特异性表达,在中脑、眼原基、端脑、菱脑、间脑和脊髓观察到清晰的杂交信号.结论: mCcd1在胚胎发育过程中的表达有阶段性,在E9和E10.5广泛表达,而在E11.5该基因表现出神经系统特异性表达,这些提示它可能参与了神经系统的发育.Objective: To analyze mCcd1 gene expression pattern during the mouse embryonic development and probe into the function of this gene. Methods:Mouse whole-mount RNA in situ hybridization of E9 to E11.5 embryos was carried out by using digoxigenin labeled mCcd1 probes and their expression profile was observed by detecting the existence and status of corresponding mRNA in the embryos. Results: mCcd1 was ubiquitously expressed at E9 and E10.5, however, it showed a neural-specific expression at E11.5. It was detected in mesencephalon (ME), optic primordium (OP), telencephalon (TL), rhombencephalon (RE), diencephalons (DI) and spinal cord (SC). Conclusion: With DIG labeling, the temporal and spatial expression of mCcd1, is examined during the mouse embryonic development. The finding suggest that it take part in the development of the nervous system because it has a neural specific expression at E11.5.
