摘 要:目的 观察维甲酸(RA)纳米粒对兔视网膜有无毒性作用。方法 30只青紫蓝兔随机分为5组:正常组,RA 5μg/ml组,RA 10μg/ml组,RA 20μg/ml组及对照组,分别于玻璃体腔注药后7、14及28 d用检眼镜观察眼底变化,然后摘除眼球,进行组织学观察,视网膜神经节细胞(RGCs)计数并测量视网膜内层厚度。结果 7 d时,RA 20μg/ml组出现 RGCs的空泡样变,但在14 d时恢复正常。各组之间RGCs数目及视网膜内层厚度无差异。结论 ≤20μg/ml RA注入玻璃体腔对于视网膜是安全的,没有对视网膜产生远期毒性。Objective To observe the toxicity of retinoic acid(RA) nanoparticles to rabbit retinal tissues.Methods Thirty pigmented rabbits(60 eyes) were divided into five groups randomly:normal group,RA 5 μg/ml group,PA 10 μg/ml group,RA 20 μg/ml group and control group.The fundus changes were observed by ophthalmoscope on the 7th,14th and 28th day after vitreous injection.The histologic changes were analyzed and the retinal ganglion cells(RGCs) were counted, and the thickness of the inner retinal layer was measured under microscope.Results RA 20 μg/ml group had bulbar degeneration in RGCs on the 7th day after injection,but recovered on the 14th day.No changes were found in other groups.Conclusion The concentration of retinoic acid nanoparticles≤20 μg/ml is safe for vitreous injection and has no long-term toxicity to the retina.
