摘 要:报道了一种从日本根酶中提取壳聚糖的简单方法。正交试验研究显示,以2%淀粉为碳源,1%蛋白胨为氮源,在起始pH5.0,26℃下培养60h时产量最高,菌丝体干重为8.43g/L。经NaOH间歇处理,HCl抽提,得到天然壳聚糖为895mg/L,壳聚糖产量占菌丝体干重的10.58%。产品壳聚糖纯度为90.5%。This paper reported a simple method for the lab-scale isolation of chitosan from hyphal walls of Rhizopus japonicus.The fungus strain was cultured with a reciprocal shaking at 170?r/min. Fungal mycelia was then harvested and the productivity determined.The orthogonal experimental results showed that culture medium with an initial pH 5.0,the optimal carbon source amylum concentration at 20?g/L,the optimal nitrogen so2酰颍悖? peptone concentration at 10g/L and temperature of 26℃ were the most suitable conditions for mycelia and chitosan production.The productivity of the dry weight of mycelia was 8.43?g/L under the optimal condition. After the mycelium was treated with sodium hydroxide twice and extracted with hydrochloric acid, we obtained the nature chitosan with a yield of 895?mg/L, which accounted for 10.58% of the dry weight of mycelia with a purity of 90.5%.
