白三烯拮抗剂ONO-1078对抗原诱导的豚鼠皮肤微血管渗漏的抑制作用

摘　　要：补尔敏预先处理的致敏豚鼠，白三烯拮抗剂ＯＮＯ－１０７８显著抑制抗原引起的依文思蓝在耳廓皮肤的渗出，静脉注射的ＩＤ５０为０．０３５ｍｇ／ｋｇ，腹腔注射为０．３６ｍｇ／ｋｇ，提示白三烯参与皮肤过敏反应，ＯＮＯ－１０７８对过敏性皮肤病变有治疗价值。但静脉注射较大剂量的ＯＮＯ－１０７８可直接引起皮肤微血管渗漏和短暂性血压降低，而腹腔注射无这些作用。ONO-1078, a leukotriene antagonist,could significantly inhibited antigen-inducedextravasation of Evans blue in auricle skin in chlorpheniramine-pretreated actively. sensitizedguinea pigs, with ID50 values of 0.035 ma/kg for intravenous administration and 0. 36 mg/kg forIntraperitoneal injection. The result suggested the involvement of leukotrienes and the therapeutic implication of ONO-1078 in skin anaphylactic response. It was also found that ONO-1078 Inlarger intravenous doses could directly elicite skin microvascular leakage and a transient hypot ention, and it had no effects while itljected intraperitoneally.
