摘　　要：目的:探讨血管紧张素Ⅱ受体 1 (AT1R)的RNAi对血管紧张素II(AngⅡ )功能的影响,我们构建了AT1R短发夹环RNA(shRNA),以期为进行这方面研究提供基础。方法:在GeneBank中查到鼠AT1R的mRNA基因序列并输入到相应设计软件中,以此设计引物序列。经PCR扩增、酶切后,克隆于T载体并行酶切鉴定。结果:构建的鼠AT1RshRNA质粒经测序鉴定验证与预期相符。结论:本研究结果为进一步研究AT1RshRNA质粒的RNAi功能及其对AngⅡ功能影响的研究奠定了基础。Objective: To investigate the function of RNA interference, the plasmids containing short hairpin RNA of angiotensin Ⅱ type 1 receptor were constructed. Methods:The sequence of rat angiotensin Ⅱ type 1 receptor mRNA was found in the Gene Bank. The primer was designed.The fragments amplificated with PCR were inserted to the pGEM~-T vector, and were selected and identified. Results:The plasmid was certified to be in the right rank. Conclusion:This study provided foundation for further research on the function of short hairpin RNA of angiotensin Ⅱ type 1 receptor and its effect on angiotensin Ⅱ.
