摘 要:研制了一个可用于对人体内血清β-羟丁酸进行定量检测的电化学生物传感器.将β-羟丁酸脱氢酶(HBDH)、辅酶烟酰胺腺嘌呤二核苷酸(NAD)等固定于丝网印刷碳电极上,以铁氰化钾为电子传递剂,采用电流法对β-羟丁酸的浓度进行测定.并对铁氰化钾浓度、NAD浓度、溶液pH值及温度等影响因素进行了优化.实验结果表明,用该传感器测定血清β-羟丁酸的浓度,快速准确、重复性好,而且线性范围可达到30~550mg/L、用血量只需2μL.β-hydroxybutyrate electrochemical biosensor was prepared to determine the concentration of serum β-hydroxybutyrate. With potassium ferricyanide as electron transfer mediator, β-hydroxybutyrate dehydrogenase (HBDH) and cofactor β-nicotinamide adenine dinucleotide (NAD) were immobilized on a screen-printed carbon electrode by direct adsorption on the electrode surface during the fabrication of this biosensor. Amperometry was used for determining the β-hydroxybutyrate concentration, and the concentrations of potassium ferricyarride and NAD, pH and temperature were optimized. The experimental results show that the response currents measured by this biosensor have good linear relationship with β-hydroxybutyrate concentration in the range of 30-550 mg/L, have high precision and good repeatability, and what is most important is that only 2 μL sample is needed.
