摘  要:为了研究分析嗜盐古生菌物种与细菌视紫红质(BR)蛋白基因资源,分离纯化得到极端嗜盐古生菌AB3. 采用聚合酶链式反应(PCR)方法对其编码螺旋C至螺旋 G的蛋白基因片断和 16S rRNA基因进行了扩增,并测定了基因的核苷酸序列. 翻译出的氨基酸序列与已报道的相应片断进行对比,AB3 中的螺旋 C至螺旋 G蛋白与其他菌株差异显著.基于BR蛋白和16S rDNA序列的同源性比较以及系统发育学研究表明,AB3 是 Natrinema属中的新成员. 菌株AB3的16S rDNA序列已被GenBank数据库收录,其序列号为AY277583.In order to study and analyze the species and bacteriorhodopsin (BR) protein resource, a strain of extremely halophilic archaea AB3 was isolated. Partial DNA fragments encoding a BR protein from helix C to helix G and a 16S rRNA encoding gene (16S rDNA) from AB3 were amplified by polymerase chain reaction (PCR), and their DNA sequences were determined. The results indicate that compared to the deduced amino acid sequences of other existing BRs, BR protein from helix C to helix G of AB3 is obviously different. The results of homology analysis on BR gene and 16S rDNA and phylogenetic analysis on 16S rDNA show that the strain AB3 is a new member of genus Natrinema. The GenBank accession number of the 16S rDNA sequences of strain AB3 is AY277583, and the research establishes a convenient and rapid method for screening a new halophilic archaeum for a new BR protein.
